Theoretically, sprites could be changed in RAM since all data is stored in RAM at some point. However, most image data is generally very large in size, and any attempts to modify that amount of RAM with a custom sprite would likely crash the game, assuming you're using Action Replay cheats or something similar.

I'm not sure if it would be useful to you, but I've seen some cheats that have successfully swapped current sprites with other sprites stored in the game.
